In uptown Manhattan, storm leaves a hanging crane and minor damage

Monday's giant storm inflicted the greatest damage on the lower half of New York City, but uptown, on West 57th Street, a massive, dangling crane is drawing crowds.

At a thousand feet above the ground, the 80-ton swing arm dangles next to the tallest condominium under construction in the city, located between 6th and 7th avenues. Bovis Lend Lease, the company managing the building's construction, is waiting for the last winds to die down before it scoops up the hanging hulk of metal.

"All I'm doing today is watching that crane," said a company employee, who asked not to be named. Wearing a muddied yellow raincoat, he leaned against a nearby shop on the corner of 56th Street, on standby.

"The only reason they haven't taken it down is because of the winds," he said. "Once they die down, they'll rig it up from the air. Right now it's no danger."

His remarks echoed the statement of the tower's developer, Gary Barnett, who told Crain's New York Business, "Everything that can be done is being done." In the meantime, metal gates and police tape block off access to the surrounding areas, as crowds of people stand along 57th Street and 5th Avenue taking photos and chattering as the crane's arm sways in the wind.

The crane is the most conspicuous damage up here resulting from the storm, but nearby, more destruction can be seen: On a building at the corner of Park Avenue and 59th Street, the facade of one floor about 20 stories up tumbled down during the storm, leaving a mass of debris on the now closed-off section below.

But for most who live around here, the most common word to describe the storm has been "boring." There have been few power outages, and the worst symptoms of the storm have been the gusts of wind. In nearby pubs like the Carriage House on 59th Street, people are hanging out, glancing at the images on TV of? more serious disorientation in lower Manhattan.

"It's honestly been a little boring," said Alana, who lives a few blocks away by the U.N. headquarters. She said she's chiefly been preoccupied with taking in her friend from Brooklyn, where winds were felt as early as Sunday night. "No power out, I've still got Internet. It hasn't been very exciting."

Others living uptown have also concentrated chiefly on coming to the aid of their friends, and checking on their work spaces downtown.

"I'm at a small tech company in Union Square, so I swung by to check on any leaking or flooding," says Brian, playing darts with a friend at the Carriage House. "Otherwise, it's mainly been working from home and helping out the people I know."

Yeast model offers clues to possible drug targets for Lou Gehrig's disease, study shows

ScienceDaily (Oct. 28, 2012) ? Amyotrophic lateral sclerosis, also called Lou Gehrig's disease, is a devastatingly cruel neurodegenerative disorder that robs sufferers of the ability to move, speak and, finally, breathe. Now researchers at the Stanford University School of Medicine and San Francisco's Gladstone Institutes have used baker's yeast -- a tiny, one-celled organism -- to identify a chink in the armor of the currently incurable disease that may eventually lead to new therapies for human patients.

"Even though yeast and humans are separated by a billion years of evolution, we were able to use the power of yeast genetics to identify an unexpected potential drug target for ALS," said Aaron Gitler, PhD, an associate professor of genetics at Stanford. "Many neurodegenerative disorders such as ALS, Parkinson's and Alzheimer's exhibit protein clumping or misfolding within the neurons that is thought to either cause or contribute to the conditions. We are trying to figure out why these proteins aggregate in neurons in the brain and spinal cord, and what happens when they do."

In 2008, Gitler received a New Innovator award from the National Institutes of Health to use yeast as a model for understanding human neurodegenerative diseases and as a way to identify new targets for drug development.

Gitler is the co-senior author of the research, published online Oct. 28 in Nature Genetics. Robert Farese, Jr., MD, a senior investigator at the Gladstone Institutes, is the other co-senior author. Stanford graduate student Maria Armakola shares co-first authorship with Matthew Higgins, PhD, a postdoctoral scholar at Gladstone.

Most cases of ALS have no clear-cut cause. However, it has recently been shown that an RNA-binding protein called TDP-43 accumulates in clumps in the cytoplasm of spinal cord neurons in many people with the condition, and mutations in this protein have been found in some people with the ALS. Researchers like Gitler and Farese have been able to mimic the disease in yeast by expressing TDP-43 at higher-than-normal levels, which causes the protein to form lethal clumps in the cells' cytoplasm.

"In humans, the progression of the disease can take years before symptoms arise," said Gitler. "But in yeast, we see protein clumping in the cytoplasm within two days and the cells rapidly begin to die." With their model system in place, Gitler and Farese set out to see whether it was possible to protect yeast cells from this effect by tinkering with the function of other proteins in the cell.

In this study, the researchers discovered that blocking the production of a protein called Dbr1 in a yeast model stops the TDP-43 clumping and allows the cells to live normally. The researchers confirmed the results in human nerve cells grown in the laboratory and in rat neurons overexpressing TDP-43.

"In this study we made no assumptions as to how TDP-43 injures cells," said Farese, "but instead screened the whole yeast genome to find genes that might prevent the toxicity. Independently, both our lab and the Gitler lab found that loss of Dbr1, an enzyme involved in RNA processing, could do this."

Dbr1 serves as part of the cellular clean-up crew that mops up the bits of unwanted RNA generated as part of the protein production line. In our DNA, most genes consist of coding regions, called exons, broken up into several segments by non-coding regions, called introns. Cells can make many different, related proteins from the same stretch of DNA by mixing and matching different exons in a process called splicing.

When the DNA is first copied, or transcribed, into RNA, the introns as well as the exons are included. But the cell quickly splices out the introns, which are released into the cytoplasm as little loops, or lariats. Dbr1, in turn, clips the loops to open them and make them accessible to the cell's disposal system.

Blocking the production of Dbr1 causes the RNA lariats to build up in the cytoplasm. The researchers showed -- by creating lariats with a binding site for a fluorescent tracking protein -- that the mutant TDP-43 binds to these excess lariats rather than clumping. The effect is like using a paper towel to mop up a spill on your computer keyboard: binding to the lariats appears to keep TDP-43 from causing havoc elsewhere.

"Normally, TDP-43 is found in the nucleus," said first author Armakola. "But in the diseased cells, it aggregates in the cytoplasm and forms clumps. We developed a novel way to track where these lariats go in living cells, and we saw that when Dbr1 is missing, the lariats act as a sink to sequester TDP-43."

The researchers note that it's still not entirely clear whether the cells die because the mutant TDP-43 is drawing essential RNA transcripts or regulatory molecules away from the nucleus and into the cytoplasm, or because it's not performing its normal RNA-binding function in the nucleus. Both could contribute to the progression of the disease.

The results in the yeast, rodent and human cells, however, suggest that therapeutic approaches aimed at blocking Dbr1 function, or even creating artificial lariat-like formations to draw away the mutant molecule, should be explored.

"Next, we'd like to explore blocking Dbr1 function in animals such as flies, worms and rodents," said Armakola. "We're also interested in identifying small molecule inhibitors of Dbr1."

Apple fails to wow as iPad sales disappoint

SAN FRANCISCO (Reuters) - Apple Inc delivered lackluster quarterly results and iPad sales fell short of Wall Street targets, pushing its stock slightly lower.

The results came as it heads into the crucial holiday season, when competition in the smartphone and tablet market will reach fever-pitch, with Apple pitting a new phone and iPads against Amazon.com Inc and Google Inc Android devices.

"We were happy with the 14 million iPad sales in the quarter. It exceeded our expectations," Apple Chief Financial Officer Peter Oppenheimer said in an interview. "But as the summer went on, the rumors were pretty rampant about the iPhone and iPad."

For the December quarter, Apple forecast revenue of $52 billion, below the average estimate of $55 billion, according to Thomson Reuters I/B/E/S.

Its fiscal fourth quarter revenue rose to $35.96 billion, roughly in line with the average analyst estimate of 35.8 billion, according to Thomson Reuters I/B/E/S.

Apple shipped 26.9 million iPhones, somewhat higher than the 25 million to 26 million that Wall Street analysts had predicted. Sales of the iPad came in at 14 million in the fiscal fourth quarter, well below lowered forecasts for the tablet.

Apple only had nine selling days of the new iPhone 5 in its fiscal fourth quarter, which means that all eyes are on the current holiday quarter.

Apple heads into the quarter after having refreshed almost all of its product lines, including an upgraded, full-sized iPad. The December quarter will show how well consumers respond to Apple's latest gamble - the new, smaller iPad mini that will goes on sale November 2.

Unorthodox Gymnastics: Why isn't rape called an abomination?

This week yet another Republican politician stepped into it--and by that I mean expressed their honest, deep seated views about rape and women's reproductive choices that are actually fairly mainstream within the leadership of the GOP. It truly sucks when you're caught on camera saying what you mean. (I'm looking at you Mitt Romney and what you think of the "47 percent.")

This time it's GOP senate hopeful Richard Mourdock who was caught saying,?"I struggled with it myself for a long time, but I came to realize life is that gift from God.?And I think even when life begins in that horrible situation of rape, that it is something that God intended to happen."

Obviously by invoking God, this raises all kinds of interesting theological questions. If God approves of the end result--the pregnancy--does that mean He also approved of the "method of conception" as Paul Ryan elegantly put it? (For the record, I'm using male pronouns for the Deity because Mourdock's God can only be male if He were to approve of forcing a woman who got pregnant as a result of rape to carry a baby to term.) And if the crime was preordained and part of "God's plan" then why can't a medical procedure such as an abortion also be part of that plan?

All of this talk of babies and rape got me wondering--the same conservatives that debate what is rape and try to classify some forms as "legitimate" or "forcible" are also often the same folks who refer to homosexuality an "abomination," as somehow unnatural and something that should be stopped at all costs. (I mean, these folks hope to pass a constitutional amendment banning same sex marriage. Thankfully, that is unlikely.) And one of arguments made against homosexual relationships is that it's not natural since gay/lesbian sex can't lead to procreation. As if the majority of sexual activity that heterosexuals engage in is procreative in purpose. (As of this writing, I have yet to engage in any sort of procreative heterosexual sex. Thank God.)

Which got me thinking about rape and the Republican weak ass, reprehensible response to it. No one calls this violent act--the violation of another person's body--a perversion, unnatural or an abomination. Is it because rape can potentially result in a baby (despite Todd Akin's protestations to the contrary)? After all, the vice presidential nominee, an opponent to gay marriage, has signified that rape, while an unfortunate occurrence, is another "method of conception." This sort of thinking allows the ends to justify the means. Even if it doesn't go that far, the fact that rape can result in a child softens some Bible thumpers' perception of it. It can't be that awful. Cause look-a baby!

And let us not forget--things like marital rape weren't considered crimes until recently and this classification still meets resistance. After all, if we're operating from a strictly biblical framework, this wasn't considered a crime. A wife was a man's property and by virtue of the contract and ceremony, her consent was considered constant. When you think about, this sort of arrangement and thinking can make rape seem "natural." As in the "natural rights" of a husband. But sex between two consenting adults of the same gender is somehow considered "unnatural."?And as we know from Republican bogeyman--science--and from visits to the zoo, same sex attraction and orientation occur naturally in nature.

So in short: baby = natural. No baby = unnatural.

If only the Bible hadn't gotten it so terribly wrong, terming homosexual acts "to'eivah" instead of placing this label on something that is actually unnatural and abominable act--rape.

Wambach, Morgan FIFA world player candidates

(AP) ? Olympic gold-medal winners Abby Wambach, Alex Morgan, Carli Lloyd and Megan Rapinoe of the United States were among 10 candidates selected for the FIFA Women's World Player of the Year.

Wambach scored five goals at the London Games, and Lloyd scored twice as the U.S. beat Japan 2-1 in the final before a crowd of 80,203 at Wembley Stadium. Morgan and Rapinoe each scored three times in London.

No American has won since Mia Hamm in 2001 and '02, the first years FIFA presented the women's award.

Christine Sinclair of Canada, who scored six goals at the London Olympics, and five-time winner Marta of Brazil also were nominated by FIFA on Thursday. Japan had three candidates including Homare Sawa, winner of the 2011 FIFA award.

Pia Sundhage, who guided the U.S. Olympic team, and Norio Sasaki of Japan were among 10 candidates for the coach's award.

Three finalists will be announced Nov. 29 in Sao Paulo after voting by national team coaches, captains and journalists.

Winners will be revealed at FIFA's annual gala on Jan. 7 in Zurich.

Dianne Bourne named 2012 College of Agriculture and Life Sciences Employee of the Year

BLACKSBURG, Va., Oct. 23, 2012 ? Dianne Bourne, a laboratory and research specialist who has worked at Virginia Tech for more than 40 years, was recently named the 2012 College of Agriculture and Life Sciences Employee of the Year. ?

Bourne, of Christiansburg, Va., supervises laboratory research in the Department of Food Science and Technology. Bourne is a leader in her department and her work was described as meticulous, scientific, and timely.

Every year the college honors a staff member who demonstrates enthusiasm and commitment, excels in professionalism, and takes the initiative to perform beyond the expectations of regular work assignments.

"Dianne employs unusual care and commitment in designing and conducting research studies," said her anonymous nominator. "The fact that our group's research is so well-respected by government agencies, private industry, and colleagues in academia is indicative of Dianne's professionalism."

Bourne critically reviews experimental designs and performs laboratory work and data analysis. She also identifies projects that are eligible for funding and assists with grant proposals.

Bourne received a $1,000 check at the college's 17th annual Celebration of the Faculty and Staff picnic.

Russell Means was one of the leaders of the Native American armed occupation of the South Dakota town of Wounded Knee, a 71-day siege in 1973. Russell Means also starred in "The Last of the Mohicans" and was guest star in the HBO series "Curb Your Enthusiasm."

Means died early Monday at his ranch in South Dakota, Oglala Sioux Tribe spokeswoman Donna Salomon said.

Means led AIM's armed occupation of the South Dakota town of Wounded Knee, a 71-day siege that included several gun battles with federal officers. AIM was founded in the late 1960s to protest the U.S. government's treatment of Native Americans and demand the government honor its treaties with Indian tribes.

Means denied the group ever promoted violence.

"You people who want to continue to put AIM in this certain pocket of illegality, I can't stand you people," Means said during an April gathering commemorating the uprising's anniversary. "I wish I was a little bit healthier and a little bit younger, because I wouldn't just talk."

Means told the AP in 2011 that before AIM, there had been no advocate on a national or international scale for American Indians and that Native Americans were ashamed of their heritage.

"No one except Hollywood stars and very rich Texans wore Indian jewelry," Means said. "That's all changed." The movement eventually faded away as Native Americans became self-aware and self-determined, Means said.

He was often embroiled in controversy, partly because of AIM's alleged involvement in a 1975 killing. But Means was also known for his role in the movie "The Last of the Mohicans" and his unsuccessful run for the Libertarian nomination for president in 1988.

Paul DeMain, publisher of Indian Country Today, there plenty of Indian activists existed before AIM, but the group became the "radical media gorilla."

"If someone needed help, you called on the American Indian Movement and they showed up and caused all kind of ruckus and looked beautiful on a 20-minute clip on TV that night," DeMain said.

Means said he felt his most important accomplishment was the founding of the Republic of Lakotah and the "re-establishment of our freedom to be responsible" as a sovereign nation inside the borders of the United States. His efforts to have his proposed country recognized by the international community continued at the United Nations, he said, even as it was ignored by tribal governments closer to home, including his own Oglala Sioux Tribe.

But others may remember him for his former organization's connection to the killing of Annie Mae Aquash, whose death remains synonymous with AIM and its often violent clashes with federal agents in the 1970s.

It?s a bit of a work in progress at the moment, but there are currently about 20 vendors listed offering an assortment of discounts and other promotions.

In particular, take a look at the company reviews in the ?Featured Services? section. These are companies I use extensively and highly recommend. Most of them are offering free services that alone would be enough for many businesses.

(AP) ? In a new sign of a widening crackdown on Russia's opposition, investigators on Wednesday opened a criminal probe against leftist leader Sergei Udaltsov and several other activists for allegedly plotting mass riots.

The Investigative Committee said in a statement it will investigate claims made in a recent documentary aired by a Kremlin-friendly TV channel that opposition leaders worked with Georgian officials to overthrow the government. But Udaltsov is not officially suspected of that more serious charge.

Udaltsov, a 35-year-old, shaven-headed Communist who wore a Stalin T-shirt for his wedding, has been one of the most recognizable faces of last winter's anti-government protests in Moscow, which were legal and peaceful.

Investigators, backed by armed men wearing ski masks, searched Udaltsov's apartment in south Moscow for more than five hours on Wednesday. The home of his parents was also searched, said Violetta Volkova, Udaltsov's lawyer.

"I'm going to hold on until the end, and I won't be quiet," Udaltsov said as he left his home for an interrogation escorted by masked armed police. "It's a wave of new repression."

A documentary aired last week on NTV, a channel seen as a propaganda arm of the Kremlin, showed what it says was footage of the Left Front leader meeting with officials from neighboring Georgia to discuss raising $200 million for protests against President Vladimir Putin, and plans for organizing riots in Moscow.

The Investigative Committee said that it would pursue criminal cases against not only Russians, but also citizens of Georgia and other unspecified countries.

"Once their involvement in the preparation of criminal acts is established, they will be subject to criminal liability under Russian law and the norms of international law, and will be issued with international arrest warrants," the committee said.

Officials in Georgia have refrained from commenting on allegations of Georgia's involvement with the Russian opposition. Lawmaker Givi Targamadze, who was featured in the NTV program as the mastermind of Georgian support, was not available for comment on Wednesday but told Georgian media last week that he had never met Udaltsov.

Udaltsov said he has met "a great number of people" recently to discuss fundraising, but all of his efforts and intentions are legal. He has insisted the footage presented in the documentary has been doctored.

The Investigative Committee said Wednesday that it had carefully studied the footage and said it was not tampered with.

Renowned human rights activist Lev Ponomarev told the Interfax news agency on Wednesday that a "broad crackdown on the opposition is very dangerous for this country" and said that early morning searches reminded him of secret police tactics in the 1930s in the Soviet Union.

The Russian Communist Party, which forms the largest opposition faction in parliament, has supported Udaltsov, dismissing allegations against him as nonsense. Party leader Gennady Zyuganov said Udaltsov is being persecuted for his views.

"The main goal is to nip the protests in the bud," he told Interfax. "There's no one left in the Kremlin who can say 'no' to that."

In addition to Udaltsov, the criminal investigation is targeting two little known Russian leftist activists, but their role in the alleged plot was not identified.

(AP) ? Teen superstar Justin Bieber has teamed up with the Adidas NEO Label to be its new global style icon.

The two-year partnership with the German sportswear company was announced Tuesday.

The Canadian pop star, who cites James Dean as his fashion idol, says the collaboration is a "perfect fit" because "we both believe in living life to the fullest, being young, free and making our own style."

The 18-year-old singer will front the company's ad campaigns and fans will be able to shop for his personally selected favorite looks. He describes his offstage style as simple, saying he chooses "clothes that look good" while onstage it's all about "comfort, great color and fit."

Bieber is touring the United States on his "Believe" tour, which moves to Europe early next year.

Russia's Prime Minister Dmitry Medvedev declared war on tobacco Tuesday, calling for a ban on tobacco advertising, an end to smoking in public and a rise in the price of a pack. Russia's smoking rates are among the world's highest, led by Kiribati where 57 percent of the adult population lights up. Here's a look at the ways some other top smoking nations are trying to bring down the number of smokers:

GREECE

Percentage of adults who smoke: 52

Since 2009, most tobacco advertising is banned except in places where cigarettes are sold, and smoking is not allowed in public spaces. The smoking ban is widely ignored, and most bars still have ashtrays on tables.

RUSSIA

Tobacco advertising is limited to marketing drives at bars and printed ads that carry health warnings. Smoking is prohibited on trains, airplanes and some other public places, but is allowed at most bars and clubs. Many cafes and restaurants have non-smoking areas.

ALBANIA

Percentage of adults who smoke: 40

Marlboro pack costs $2.20

Tobacco advertising is banned in Albania, and the law prohibits smoking in bars, offices and other places where people gather, but the rules are sometimes flouted.

BULGARIA

A program introduced last year to limit tobacco advertising is commonly ignored, with celebrities commonly appearing on billboard ads. As Research In Motion continues to lose market share in the United States, some users still on their BlackBerry are reportedly embarrassed. Well, that's at least according to The New York Times.

In a piece called "The BlackBerry as Black Sheep," some users express a bit of shame for having to continue to use a BlackBerry.?

"I want to take a bat to it," Rachel Crosby told the Times. "You can't do anything with it. You're supposed to, but it's all a big lie."?

While this is obviously not a scientific survey, it's clear that RIM's market share and reputation have taken a big hit over the last few years as the iPhone and Android devices have grown in stature. At the same time, there are still a lot of people who have to use a BlackBerry for work -- that's starting to change, though.

RIM wasn't thrilled with the article or the fact that the Times didn't reach out to it before publishing the piece.?

US Treasury Department postpones foreign exchange policy review ...

Markets Overnight

The Chinese data released since Friday have been encouraging but this has not been rewarded by the market. China?s exports in September beat expectations increasing 9.9% y/y (consensus 5.5%) from 2.7% y/y in August, see Flash Comment China ? Encouraging exports but imports remain subdued, 14 October. It does not change our view that China?s exports will face headwinds in the coming months but the September data suggest that weak exports should weigh less on growth in the current quarter. Growth in money supply M2 in September also accelerated faster than expected to 14.8% y/y (consensus 13.7% y/y) from 13.5% y/y in August, see Flash Comment China ? Credit and money supply expanding faster, 14 October. Data released Friday also showed stronger growth in broad credit measures, albeit the bank?s local currency loans came in slightly below expectations. Finally, CPI inflation in September eased to 1.9% y/y in line with expectations from 2.0% y/y in August, mainly on the back of lower food prices, but this will be reversed in the coming months when we expect inflation to climb above 2.5% y/y on the back of higher food prices. These data have eased the pressure for imminent monetary easing in China, even though the GDP and industrial production data released later this week are expected to remain subdued.

Late Friday evening the US Treasury department announced that the publication of its bi-annual review of its major trading partners exchange rate policy will be delayed until after the US presidential election. The report was scheduled for release today but has become a hot potato in the election campaign with Republican candidate, Mitt Romney, promising to label China a currency manipulator if he is elected. The IMF and World Bank annual meeting closed this weekend and left the impression that there is no clear international consensus on how to move ahead. The Spanish Finance Minister, Luis de Guindos, at a press briefing in Tokyo did not indicate Spain is in a rush to ask for ESM assistance.

There is a slightly negative risk sentiment in the market this morning. US stock market closed moderately lower and this morning most Asian stock markets are also moderately lower, however with Nikkei bucking the trend. US bond yields have edged slightly lower since market closed in Europe Friday. In the FX market EUR/USD has declined 0.4% since Friday afternoon while the Scandinavian currencies are trading largely unchanged. Brent crude oil is 0.6% lower on possible reopening of negotiations with Iran.

Lauren Eckman, the 2013 Georgia Teacher of the Year, will speak at 7 p.m. Thursday (Oct. 18) in McAllister Hall auditorium at Berry College.

Eckman?s speech, ?Imagine the Possibilities,? will share her philosophy and practice as she works to reach every child and empower them to succeed. She is a teacher of the blind and visually impaired at the Georgia Academy for the Blind, where she teaches high school English language arts and transition courses. She works diligently to empower her students to achieve their highest levels of independence by aiding them in the acquisition of reading, writing, research and analytical skills in the traditional curriculum.

Eckman assists her students in the development of self-advocacy, as well as social and daily living skills in the Expanded Core Curriculum, which is a prescription of specialized courses designed to provide compensatory skills to individuals with visual impairments. This event is free and open to the public.
